What are NL-PEPTIDES?

NL-PEPTIDES™ are innovative analogues of classic peptides—designed to maintain their biological activity despite the effects of stomach acids, digestive enzymes, and high temperatures.

Thanks to precise modifications of the chemical structure (acetylation and amidation of the amino acid chain ends), we have obtained peptides with stability comparable to the body's natural proteins.

Their amino acid profile remains intact, meaning they retain their original shape and full activity even after passing through the digestive system.

As a result, NL-PEPTIDES™ are effective not only after injection but also when taken orally, something previously impossible with traditional peptides.

What is NL-PEPTIDES-DELIVERY™?

Patented double-capsule technology for effective peptide delivery. NL-PEPTIDES-DELIVERY™ is our patented oral peptide delivery system.

It was developed to address two of the biggest challenges faced by peptides: degradation in the stomach and poor bioavailability in the intestine. The outer capsule is coated with a special acid-resistant coating, allowing it to bypass the stomach and reach the small intestine intact.

Inside, there's a second capsule—a microcapsule containing the active peptide, a protease inhibitor (a substance that blocks enzymes that break down proteins), and an absorption enhancer that facilitates the compound's penetration through the intestinal barrier. This technology ensures the peptide is fully protected until absorbed and can act with full biological effectiveness.

In vitro and in vivo study results clearly confirm that NL-PEPTIDES-DELIVERY™ increases plasma peptide concentrations several times over conventional capsule forms.

What problems did we solve?

For years, efforts have been underway to develop a way for peptides—extremely promising biological compounds—to be effectively administered orally. Problems have included the molecules' instability, degradation in the stomach, and very low bioavailability. At Synthagen Laboratories, we have developed technologies that completely eliminate these limitations.

Peptide instability

Why this is a problem:

Classic peptides are extremely sensitive to light, temperature, and moisture.

Under these conditions, their amino acid chain rapidly breaks down, causing them to lose biological activity and effectiveness.

Our solution:

By developing NL-PEPTIDE™ analogs – structurally modified forms with increased resistance – we have obtained molecules that are stable in the external environment and more resistant to the action of digestive enzymes.

Each of our peptides retains full biological activity and durability, even in conditions that would be destructive for classic forms.

Degradation in the stomach

Why it's a problem:

The acidic environment of the stomach (pH 1–2) breaks down classic peptides before they reach the intestine.

As a result, most oral forms available on the market have no real effect.

Our solution:

NL-PEPTIDES-DELIVERY™ technology is a patented double capsule system. The outer layer is resistant to hydrochloric acid and allows for safe passage through the stomach, protecting the contents from degradation.

Only in the small intestine does the controlled release of the active peptide occur in an environment conducive to its absorption.

Low bioavailability and limited absorption

Why it's a problem:

Even if the peptide survives the digestive process, only a small fraction of it has a chance of crossing the gut-blood barrier.

Without additional support, most of the active substance is simply excreted.

Our solution:

Inside the NL-PEPTIDES-DELIVERY™ capsule there is a protease inhibitor (blocking protein-digesting enzymes) and an absorption enhancer that facilitates the penetration of the peptide through the intestinal wall. This solution allows for real oral bioavailability, confirmed by tests of peptide concentration in the blood within the range of 30–360 minutes after consumption.

Stability of peptide lyophilisates during storage.

Lyophilized samples were stored in sealed vials for 365 days at 35°C and standard humidity. Analyses were performed by opening each vial at monthly intervals.

Stability of aqueous solutions during storage.

Structural stability was assessed by analyzing peptide solutions prepared as follows: Peptide solutions were prepared at a concentration of approximately 1 mg/ml in demineralized water (pH approximately 5.6). These solutions were stored under controlled conditions at 40°C and 55% humidity, and assays were performed at regular intervals.

Results of tests for NL-peptide concentrations in blood

The first three example graphs show peptide peaks from properly prepared blood samples, analyzed over a 120-minute period. For NL-GHK-CU, NL-EPITHALON, and NL-BPC-157, administered using our patented NL-PEPTIDES-DELIVERY™ delivery technology, and for the standard peptides GHK-CU, EPITHALON, and ARG-BPC administered in standard capsules. The next three graphs show the distribution of peptide concentrations in the tested blood over a time period of 30-360 minutes, converted to pg/ml.

Conclusion: Our dual-encapsulation technology makes oral peptide administration possible. The graphs below demonstrate how our technology works.

BIOTECHNOLOGICAL RESEARCH - how do our products work?

For example, we have conducted studies such as: assessing the impact of in vitro digestion on active ingredients in products, including assessing the bioavailability of active ingredients across the gut-blood barrier. We also assessed the rate of release of active ingredients (peptides) from products in the gastrointestinal tract, as well as determining dosages, and much more.

Conclusion: Our NL-peptides penetrate the intestine intact and easily cross the gut-blood barrier. Simply put, they work, and their amino acid profile remains unchanged.

PRODUCT COMPOSITION RESEARCH - WHAT AM I BUYING?

The tested samples were applied in 15 µl amounts to a Thermo Scientific BioBasic 8 column. During the linear gradient analysis, peaks of the desired substances appeared in H2O and acetonitrile solutions with 0.1% trifluoroacetic acid added. Their concentrations, after comparison to the reference chromatogram, are presented in the tables. The measurement error is +/- 0.5 mcg.

Conclusion: Our products contain the composition consistent with the declared content.
